FOXO4-DRI is a synthetic, cell-penetrating peptide designed to disrupt the interaction between FOXO4 and p53 inside senescent (“zombie”) cells. It’s built using D-retro-inverso (DRI) chemistry — amino acids in reverse sequence with D-isomers substituted for L-isomers — which resists normal enzymatic degradation and extends the peptide’s stability in research settings.
Research focus: Research centers on senolytic activity — selectively inducing apoptosis in senescent cells while sparing healthy proliferating cells — by displacing p53 from the nucleus of senescent cells and disrupting FOXO4’s protective interaction with it. Preclinical studies have examined this mechanism in models of aged/damaged tissue (including chondrocyte and fibroblast senescence models) and in models relevant to fibrosis and chemotherapy-induced senescence.
